原标题:来自裙带菜孢子叶的高分子量岩藻多糖免疫调节功效

①免疫调节包括免疫刺激和免疫抑制,免疫刺激剂可以缓解肿瘤化疗引起的副作用,打破体内肿瘤免疫耐受;
②岩藻多糖是来自于褐藻中的含硫酸基的多糖物质,包括岩藻糖、半乳糖等,具有广泛的生物活性,包括增强免疫力、改善胃肠道及修护肾脏等作用;
③本文利用体内和体外实验,评价了来自裙带菜孢子叶的高分子量(258.7kDa)和低分子量(54.1kDa)岩藻多糖增强免疫效果;
④体外实验结果显示,相同浓度下,高分子量岩藻多糖对促进自然杀伤细胞(NK细胞)增殖及其活性提升具有更好的效果;小鼠体内实验结果显示,低分子量岩藻多糖能将NK细胞活性从63%提升至70%,高分子量岩藻多糖能提升至76%;同时高分子量岩藻多糖能促使小鼠体内产生更多的白细胞介素-2、12(IL-2、12)、干扰素-γ(IFN-γ)等细胞因子及颗粒酶,免疫刺激效果更好。
⑤结论:来自裙带菜孢子叶的高分子量(258.7kDa)岩藻多糖能够显著增强NK细胞活性,促进小鼠脾脏内细胞因子(IL-2、IL-12、IFN-γ等)分泌,相比低分子量(54.1kDa)岩藻多糖具有更好的增强免疫效果。

Characterization and Immunomodulatory Effects of High Molecular Weight Fucoidan Fraction from the Sporophyll of Undaria pinnatifida in Cyclophosphamide-Induced Immunosuppressed Mice
Abstract:
Immunomodulation involves two mechanisms, immunostimulation and immunosuppression. It is a complex mechanism that regulates the pathophysiology and pathogenesis of various diseases affecting the immune system. Immunomodulators can be used as immunostimulators to reduce the side effects of drugs that induce immunosuppression. In this study, we characterized the chemical composition of high molecular weight fucoidan (HMWF) and low molecular weight fucoidan and compared their functions as natural killer (NK) cell-derived immunostimulators in vitro. We also tested the effectiveness of HMWF, which has a relatively high function in vitro, as an immunostimulator in immunosuppressed animal models. In these models, HWMF significantly restored NK cell cytotoxicity and granzyme B release to the control group level. In addition, the expression of interleukin (IL)-1, IL-2, IL-4, IL-5, IL-12, interferon (IFN)-γ, and tumor necrosis factor (TNF)-α also increased in the spleen. This study suggests that HMWF acts as an effective immunostimulant under immunosuppressive conditions.
